Very cool new approach to tackling viruses

"If you look at the viruses that are the biggest threats of modern times, most of them were unknown through human history: HIV, SARS, Ebola (see A spotter's guide to human viruses). You don't know where the next one is coming from. How do you develop therapeutics for the unknown and unknowable, given that you won't have time to develop a vaccine for a new agent after it appears?" he asks.

Goldblatt and a few other researchers think they have the answer. They are working on an entirely new class of antiviral drugs that should do something seemingly impossible: work against a wide range of existing viruses and also be effective against viruses that have not even evolved yet. What's more, it should be extremely difficult for any virus to become resistant to these drugs.

Back in the late 1990s, when Goldblatt was at DARPA, he began to wonder whether there was another strategy, one that exploits the key weakness of all viruses: their utter dependence on their hosts. By themselves, viruses are more helpless than newborn babies. They can replicate only by tricking their host cells into making more copies of them, a process that can involve hundreds of host proteins.

What if, Goldblatt wondered, some host proteins are essential for viral replication but not for the survival of the host? If so, disabling these proteins should block viral replication without killing healthy cells.

After moving to Functional Genetics, Goldblatt began putting his idea to the test. He and his colleagues disabled one gene at a time in human cells before exposing them to viruses such as flu. This fishing expedition worked beautifully: they identified more than 100 different human proteins that flu viruses need to replicate but which cells can survive without. Only four were previously known to be involved in viral replication.

One especially promising target is TSG101, a protein involved in the transport of materials within cells that many viruses co-opt to break out of cells. Functional Genetics has developed a small-molecule drug that appears to block the interaction between viruses and TSG101. Dubbed FGI-104, the drug inhibits a wide range of viruses in cell culture, including hepatitis C and HIV, and has also been shown to protect mice against Ebola (American Journal of Translational Research, vol 1, p 87).

There's a long road ahead, and it's not yet certain that it'll work, but holy shit that's a good idea.

Cool, yes. But I too wonder about the unintended consequences. You've got to think those other proteins are there for a reason or else they wouldn't be there. They may not have an immediately noticeable purpose, but it's hard to believe they are completely superfluous. Afterall, God put them there for reasons only he knows.

Me too but I wonder what happens to those proteins that got hijacked into thinking they were doing the body good? If they get killed anyway what's the big deal. Even better though....

Since we know the specific proteins it is feasible that we create new proteins that replace the problem proteins in function but are engineered to be virally resistant.

I bet they have thought of your concern and that's why the road still has some bumps to work out.

You know... my first time here didn't go so well. I got really frustrated.

This time around I'm actually not getting stupid and agitated.

So if people can let someone start over then I'd like that. If not then well... I don't know.

I like to climb. The people here are great people and I messed up. I want to make a lot of excuses and say this or that but that would be kind of lame wouldn't it.

As rockreaver I'm myself. As codefrog 4 people had the ability to sign in as him and they did. I've tried to get codefrog unbanned because I think that a person should be able to say "I messed up. Sorry about that." and start over.

A few people here have known all along who rockreaver was and it hasn't really bothered them. I debated loading that picture but then I thought about more and realized that if you have to live a lie to be accepted by people then it ain't worth it.

Codefrog is still banned. I'm the only person who is/was rockreaver and I'm not giving that sign-in to anyone.

Codefrog was a guy from france, a guy from India and 2 guys from Australia. Rockreaver is just me.

I just want to climb, learn, be humble and not have to duck or hide my face.

So is that okay?

{EDIT} Your comments are a bit depressing though. I've lost 30 pounds between photos and you didn't notice. I'm not in the Navy. I'm not a SEAL. I'm not any of that military stuff either. I have some friends who are and that's about it. When I gave up the profile information the 4 guys decided to be 4 faces and it rolled from there. BLONDGECKO stopped it all and I think I took a bit of time off here. Now I'm back. {/EDIT}

Nope that's true. I swear on all things holy that's true. 4 guys I write software and have known for 9 years. They are beer drinking trolls and vikings and I swear to GAWED they collaborated on every post. I never in my life thought I'd be back here but this is a great place and my kids are into climbing a lot.

Those 4 vikings are devious and they plotted to be me and I'm not shitting you at all. I swear. They know me well. I'm extremely verbose and I say too damned much. They made sure to be the same. I swear.

I was a senior engineer for Micron Technology and wrote their backlog management system for memory modules and wafers. I wrote the Wildlife Management System for Idaho Fish & Game. I wrote the archived license lookup system for Idaho Fish & Game.

When it comes to source code I'm really good. I just got annoyed and on an impulse gave the account away. Then it got worse and worse. I followed it for a bit and then just kind of got really ashamed. So I tried to come back as wayfarer but somebody spotted that in no time flat and I was on the defensive so I gave up.

I registered rockreaver but I didn't post for a very long time (or I think that's right... that was the plan). Then I just tried to be very vanilla. I had real questions I wanted help with and I knew there were good people here. Some of them whom I won't mention have been cool since day one and each time I morphed I told them that I was agent X or Y or Z and they were like "okay".

I have been paid very well to catch hackers. I've programmed CISCO IOS for firewalls. I've done it all and I'm pretty good. I'm pretty burned out on life though keeping a kid alive and the debt has me really fried. All I want to do is climb. That means I hang out here and I read, and read and read and read and read.

Then I add a picture and my whole world goes right to hell. Oh well. You get what you earn and I earned what I'm getting.

A few days ago I signed in as codefrog and was still banned but I forgot so I thought that I had finally irritated the hell out of BlondGecko and maybe he banned me. So then I signed in as Wayfarer to see if it would work and it did work. When I say work what I mean is that there were buttons to reply and stuff. For codefrog I cannot view forums at all so posting is definitely out. Wayfarer can do everything but aside from signing in I haven't used it and neither has Dingus.

In hind-sight I think that banning or deleting wayfarer is good and keeping codefrog banned is good too. There's just too much drama there. I wanted BlondGecko to unban it so I could apologize with it and that's all.

I intended to stay RockReaver forever because I was or seemed to be building a presence here that had credibility that was starting to come together.

I am still blown away that he recognized me. The pictures are so different. Man!

So at this point I'll accept whatever judgement you want to pass. But RockReaver is the real deal. I'm not hear to jack people around. Not at all.

- Delete wayfarer and codefrog though if you would like. I don't think I'll use them ever. I just like being RockReaver. An okay 5.9, wobbly 5.10 and shitty everything else climber with 3 kids and a great wife. That's all I want to be. I'm not some SEAL (that was absurd) and I'm not some military pony-boy either. Just a vanilla dude.

I think what sets me apart is that I've lost 100 pounds and managed to hold a family together after having a child nearly die a lot over 3 years, losing a job and losing a house. Nothing really special there but those are the only things I've accomplished in life. Not a lot but it's at least stuff to be happy about. When I lead my first 5.10 climb I'm going to be happy as hell. Then I'll feel like I'm getting somewhere.

See here I am *verbose* as hell. I can type really, really fast. Not that it's a bragging point.

So anyway I just want to be a normal joe. Trying to do his part and climb.

I never said anyone broke into anything (I don't think.) and I didn't set up Wayfarer to just give it to Dingus. I was trying but blew it and I thought... Crap. Maybe if Dingus plays around it'll lighten the mood and this will blow over. It didn't at all. So I stopped after that.

Respecting my posts. I'm not going to sit here and ask you to respect anything. I guess each of you have to decide what you want to do. But the real me (rockreaver) never once made a post that wasn't sincere. Never.

I thought I came across pretty honest but whatever. I'm at the point where this isn't really about me anymore. You guys know who I am.

Shouldn't I be a little frustrated? 150+ posts over a few months and being largely accepted by the norms. Then I add a profile picture and now I'm right back to where I was.

I don't know. Seems like that would get a few people kind of frustrated.

Well anyway... 3 months give or take... yeah a little frustrated.

{EDIT} Your comments are a bit depressing though. I've lost 30 pounds between photos and you didn't notice. I'm not in the Navy. I'm not a SEAL. I'm not any of that military stuff either. I have some friends who are and that's about it. When I gave up the profile information the 4 guys decided to be 4 faces and it rolled from there. BLONDGECKO stopped it all and I think I took a bit of time off here. Now I'm back. {/EDIT}

I was a senior engineer for Micron Technology and wrote their backlog management system for memory modules and wafers. I wrote the Wildlife Management System for Idaho Fish & Game. I wrote the archived license lookup system for Idaho Fish & Game.

When it comes to source code I'm really good. I just got annoyed and on an impulse gave the account away. Then it got worse and worse. I followed it for a bit and then just kind of got really ashamed. So I tried to come back as wayfarer but somebody spotted that in no time flat and I was on the defensive so I gave up.

I registered rockreaver but I didn't post for a very long time (or I think that's right... that was the plan). Then I just tried to be very vanilla. I had real questions I wanted help with and I knew there were good people here. Some of them whom I won't mention have been cool since day one and each time I morphed I told them that I was agent X or Y or Z and they were like "okay".

I have been paid very well to catch hackers. I've programmed CISCO IOS for firewalls. I've done it all and I'm pretty good. I'm pretty burned out on life though keeping a kid alive and the debt has me really fried. All I want to do is climb. That means I hang out here and I read, and read and read and read and read.

Then I add a picture and my whole world goes right to hell. Oh well. You get what you earn and I earned what I'm getting.

Just to harp on this point a bit more (man, I hope I don't derail the promised goat-tit pictures), which was it? 4 guys you know really well, and a "go have fun;" or an "I got annoyed & gave it away?"

Anyway, are the g-t's of similar quality to Lucy's?

It was 4 guys that I know well that are laughing their asses off right now. Here's how the conversation went. I screw around with people a lot. So things like, "Guys here's a login to this web account. Go have fun. Come pretty naturally for me. Plus the 4 guys I've known since college take fun pretty seriously.

I'm not lying when I say this. We once carried a cement parking spot thingie up 4 flights of stairs and propped it on someone's door. Dumbest thing we ever did. Nobody got hurt but splitting $1000 5 ways in college to repair the apartment is a bit hard.

We drove a golf cart off an embankment and flipped it (totally on accident and I had nothing to do with it but I was there). We filled up all the holes on the course with sand. These guys would carry empty beer bottles around with them so they could swap them for fulls at bars when people weren't looking. Then all night long they'd swap the empty. Look! Free beer.

The list of exploits goes from there So this came pretty naturally. All of these guys are American too but they hail from their homeland. There should be a french guy in their too from an exchange program but he was on vacation (he's always on vacation).

What on earth did you guys learn in college? I mean I'm in zero position to judge here but I got a masters in jacking people around. The list of exploits would make a great book. Anyway, it's pretty much out of the bag now. I cannot convince anyone of anything. My phone number is all over the web get it and call me and you'll find out I'm not anything more than a 35 year old smart ass that enjoys life a lot and made a mistake here but aside from that has a pretty decent track record with humanity.

Pranks! I swear that was 101 in college but maybe it's just me. The guys I ran with had a ton of fun. The police knew us all by name and even liked us. We never did *real* crime but we kept them on the beat. We've actually bought police dinner and let them quiz us about our exploits and they laughed harder than we did. One of them gave us his glock which we promptly took all his bullets out of until he paid for dinner. Stuff like that was a normal thing. I was a bike mechanic for about 6 police officers and I'm pretty sure they enjoyed the hell out of us.

But in terms of "thinking this up" that was cake. That came pretty naturally.Angelic

Living vicariously through what? You have to think about this. What vicarious personality am I living right now? If I am it's pretty dull. Wouldn't I have picked something more interesting? I mean I've been asking about trad harnesses, soft shells and slings. That's not much of a vicarious life. Not really at all.

In real life I'm self-employed and I do very well. I know most of the major businessman in this town as well as the CEO of the largest memory maker in America. Just saw him in McCall a few weeks ago and we both talked for about 30 minutes while hanging out on a dock at the waterfront of a lake.

If I'm living vicariously I'm not doing a very good job at all. As far as hating violence mmm not so much. It has it's place and I'll leave it at that. I have good friends in Afghanistan and Iraq as well as other places in the world. Good people and they are... very violent if the situation mandates it. But they are officially "peace keepers" and they like that too. They want to come home to their wives and kids too.

Okay. I'm not really going back into that explanation. No I'm not still doing. It was a mistake and one I won't make again but I think I've said enough on that. It'd be pretty hard to explain this to my kids and the fact that I'd have a hard time explaining it means it was a bad idea. A really bad idea.

But if you look at the events I wasn't really wanting this to happen.

Things were going just fine as rockreaver. I wasn't trolling. I wasn't yanking/wanking. I was just hanging out and getting my feet down. I added a profile picture and Burns was on it. I knew that it was going to collapse from there and it has.

I can restate the whole story for clarity or not. I'm out. I ruined the thread on accident. Well I just changed my profile pic and the thread collapsed after that.

- I explained the situation.
- I admitted it was a mistake.
- I apologized.
- I'm trying to make it right.

Can we wrap this up? I don't know what you guys are really after now. If you don't like me then ignore my posts. But this is now getting iterative. We are going back to places we've been and in going there it's not really doing much. Have the mods ban me. Wait you are a mod.

* If you think that anything rockreaver has ever done deserves banning then do it.
* If you think that rockreaver is still pulling pranks then ban him.
* If you think rockreaver is intentionally doing things to interrupt the purpose and intent of this site then ban him.
* If you cannot let someone have a 2nd chance then ban me.

I guess I want to say this. Judge me by my actions as rockreaver both now and moving forward. Can we let the rest of this fall away? I just want to be a climber that is learning the sport. I'm also getting into snow shoes and winter camping. I want my son to have confidence and a good work ethic in life.

None of this would even be happening if we were face to face. You'd have either punched me and walked away. You'd have said, "It's fine dude. Just don't do it again. Buy my lunch, belay me on anything I say and we'll forget this happened." or you'd have laughed it off.

This is dragging out because it's the web. Drive over and punch me. Make me be your sherpa or forget about it. I've done everything I can do. Short of sending you all a some of Dingus's porn there's not much left to do.

I'm not going to make this a whole lot of fun. There's information I need here. There are relationships I have here. There are things I need to learn here. So either ban me or let it go.

I'm outta here though. I just picked up a Cloudveil Rayzar for $227 at Sierra Trading Post and an Icebreaker 260. Sierra Trading Post did something really cool. They honored a 20% off coupon that I just got on the jacket that I bought 5 days prior but they further applied it to the Icebreaker so the Icebreaker costs me $6. Now I need to figure out what outer parka would be a good match for that and then I'll be outfitted for winter climbing and snowshoe trekking.

I have to move on. This is never at all what I intended and sometimes you do things you regret. All of us do. You just want to start over and leave it behind.

Codefrog is dead. Wayfarer is dead. Rockreaver is humbly working his way into a long life of climbing and outdoor activity and if the few of you here are okay with that I'll be moving on.

I promised my son a climbing tour when he graduates from high school that will finish at Yosemite and hit other places like Squamish and possibly the Tanks. We'll have fun.

Someday I'll tell him this story when he's old enough to understand it and he'll probably look at me and say, "Dad that was a dumbass thing to do." and we'll agree and keep driving.

I'm off guys. Lock the thread, give it back to BlondGecko or something. I've got a normal life to live not a past to be mired in.

{EDIT}The reason I changed my profile pic is that yesterday marks the day I got a membership at the YMCA. I weighed 270 pounds. I'm now 170 pounds and to celebrate that I took a photo and am using that photo on all my profiles. What if, Goldblatt wondered, some host proteins are essential for viral replication but not for the survival of the host? If so, disabling these proteins should block viral replication without killing healthy cells.

After moving to Functional Genetics, Goldblatt began putting his idea to the test. He and his colleagues disabled one gene at a time in human cells before exposing them to viruses such as flu. This fishing expedition worked beautifully: they identified more than 100 different human proteins that flu viruses need to replicate but which cells can survive without. Only four were previously known to be involved in viral replication.

One especially promising target is TSG101, a protein involved in the transport of materials within cells that many viruses co-opt to break out of cells. Functional Genetics has developed a small-molecule drug that appears to block the interaction between viruses and TSG101. Dubbed FGI-104, the drug inhibits a wide range of viruses in cell culture, including hepatitis C and HIV, and has also been shown to protect mice against Ebola (American Journal of Translational Research, vol 1, p 87).

There's a long road ahead, and it's not yet certain that it'll work, but holy shit that's a good idea.
